Dario Rosso
Voiced by: Tony Rosato (1999) and Rick Zieff (2020), Binbin Takaoka (Japanese).
A salesman who was with his family in Raccoon City when the outbreak began.
- Adaptational Jerkass: In the remake, Dario makes no mention of his family when encountered, and is instead presented more as a bitter, selfish individual who only cares about himself, actively telling Jill to go and get herself killed when she tries to take him to safety. He's such an asshole that the Record (an in-game challenge) for speaking to him multiple times is even titled "Fuggedaboutim!"
- Age Lift: He looks considerably older in the remake, not to mention sounds and acts even more curmudgeonly than before.
- Asshole Victim: He's enough of a Jerkass to Jill that most players won't be particularly saddened by his death. Bonus points in that even his last diary entry before his death seems self-absorbed and arrogant.
- Jerk Ass Has A Point:
- For all of his paranoia and hostility toward Jill in the remake, he turns out to be correct in being safer locking himself in the warehouse than going with Jill when Nemesis shoots down the helicopter and nearly kills her.
- In the original, he expresses how dangerous it is outside the warehouse and states that he's safer in the container. Shortly after leaving the container he immediately gets eaten by zombies, kind of proving his point.
- Sole Survivor: By the time Jill finds him in a warehouse, he is the only survivor of his family and this doesn't last long.
- Too Dumb to Live: Although he is hysterical with panic, he still has shades of this. First of all he keeps screaming at the top of his lungs that he doesn't want to be eaten by zombies, ignoring the fact that this will probably attract them. He also angrily refuses to follow Jill, even though she's perhaps the only person trained and qualified enough to get him out of the city alive.
- Tragic Dream: Early in life, he wanted to be a novelist but was persuaded by his mother to become a salesman instead to support his family. With his family dead and now trapped in a city filled with monsters, Dario deeply regrets never having written a book in his life.

Daniel Fabron
"Bonjour, mon ami! The wheels of fate led you to a strange place today, non? Let us see if you walk away...or not."
Voiced by: Kaiser Johnson (English), Hiroshi Shirokuma (Japanese)
Spencer's personal "fixer" and Alex Wesker's assistant. Though he isn't a scientist, he is well-versed in controlling B.O.W.s. His playstyle focuses on summoning and controlling powerful creatures, including the devastating Tyrant.
- Large Ham: He uses the intercom to broadcast grandiose taunts and boasting at his victims constantly.
- Laughably Evil: Daniel taunts his victims to such an audiacious degree that he comes across as a cocky little gamer kid.
"Mine, mine, mine! Hahaha, fuck you all!"
- The Nicknamer: He is very fond of giving pet names to B.O.W.s
"I call this one The Poisoninator! ...eh, the name could use some work."
- Original Generation: Among all of the Masterminds, he is the only character created specifically for Resistance.
- Signature Mon: His bioweapon is the T-103 Tyrant, more colloquially known as "Mr. X", who assails his victims with powerful punches and deadly grabs.
- Sir Swears-a-Lot: He puts on an air of suaveness when things are going well. When they're not, he'll start swearing up a storm.
- Sore Loser: He doesn't suffer defeat very well.
"What is this motherfucking bullshit?! Dr. Wesker will think I barely tried!"
- Villain Override: His playstyle favors not only summoning monsters in large numbers, but also taking control of them and giving them powerful buffs while under his command.
Survivors
Samuel Jordan
A professional boxer who was forced into retirement due to an injury and joined in Umbrella's research in the hopes of restarting his career. Samuel is a close-range bruiser who can use his boxing skills to defend himself, using Dash Punch to charge down enemies and Fists of Iron to beat them down.
- Bare-Fisted Monk: While he can use guns and is handy with melee weapons, he is also more than capable of fending for himself with his bare hands.
- Dumb Muscle: While Samuel isn't stupid by any means, he is one of the most combat and melee-oriented survivors, and most of his voice lines makes him come across as a Dumb Jock.
- Healing Factor: His Fever Skill and his Dash Punch have unlockable abilities that grant regeneration.
- Super-Soldier: Perhaps as a result of the experimental treatment, it grants him enhanced strength and a limited healing factor.
January Van Sant
A hacktivist who was abducted by Umbrella after looking into their activities and their connections to the Raccoon City Police Department. January is a saboteur who can use her hacking abilities to interfere with the Mastermind, using Overload to temporarily disable cameras and EMP to interfere with all cameras while increasing the Mastermind's cost to use their abilities.
- Defrosting Ice Queen: The game describes her as a lone wolf who will do anything for money, and her comments to masterminds tend to be highly aggressive. Her victory quotes usually have her praising her team, or being pleasantly surprised at how well the team worked together.
Valerie Harmon
A grad student and expert chemist who was hired as an intern by Umbrella, then subsequently abducted when she started looking into a roommate's memory disorder. Valerie is a supportive character who can heal her allies personally with her Modified First-Aid Spray, as well as more easily identify threats and items with her Survival Instinct.
- Combat Medic: She serves as the primary healer among the Survivors. Her Modified First-Aid Spray ability creates an Area of Effect that heals all Survivors in range.
- Spider-Sense: Survival Instinct allows Valerie to detect enemies and useful items.
Tyrone Henry
A firefighter with the Raccoon City Fire Department who was abducted while battling a fire at an Umbrella facility. Tyrone is a tank who can keep enemies at bay with a Power Kick, as well as Rally his allies to help them take less damage.
- Big Damn Heroes: His kick can save other survivors from the grip of a Bioweapon's instakill move, something anyone else needs an explosive to do.
- The Leader: The masterminds tend to comment on how the other survivors look to him for leadership, and all of his dialogue is both confident and encouraging to the team.
- Stone Wall: By using Rally, he can decrease the damage he and nearby survivors take for a brief time. In comparison to other survivors, however, his offensive capabilities are not as spectacular: he can get some breathing room with Power Kick, but that's about it.
Becca Woolett
A park ranger who was abducted by Umbrella following an encounter with mutated dogs. Becca is a sharpshooter who is well-versed in firearms: her Sentry Stance improves her aim considerably, and with the Bullet Storm ability, she can fire without spending any ammo for a short time.
- Ms. Fanservice: She's an attractive blonde, and wears the most revealing outfits out of everyone, consisting of shorts and mini-skirts, which all bare her midriff.
- The Gunslinger: Guns are Becca's forte. Her Sentry Stance makes her deadlier with firearms, and Bullet Storm gives her infinite ammo for a short time.
Martin Sandwich
A mechanic who stumbled upon Umbrella secrets while conducting repairs at Spencer Memorial Hospital, secrets that some would prefer not be known. Martin is a gadgeteer who can build his own traps to keep B.O.W.s at bay, such as a Makeshift Mine to blow creatures away, or a Flash Baton to stun them.
- Gadgeteer Genius: He's able to cobble together functioning and deadly traps out of assorted random junk that he collects from around the environment.
- He Knows Too Much: He wasn't even trying to uncover Umbrella's dirty secrets. Merely stumbling upon them made him fair game for Umbrella to take out.
